I would suggest trying a 1/2 hour first, simply because that bike seat is VERY uncomfortable at first! Or perhaps do 2 30-minute segments in a day. Pick a resistance that is easy for a warm up, and after a few minutes, pick a resistance that is challenging but not incredibly difficult to start.
You can do an endurance ride, which would be riding at one resistance/speed for the entire time, or you can do intervals to spice things up. The intervals can be speed (30 seconds pedaling as fast as you can, then 2 minutes to recover, etc.) or resistance (30 seconds at a higher resistance, 2 minutes at an easier recovery resistance) and the interval times can be changed to suit your fitness and energy levels.
I agree with Jules, that seat is very uncomfortable, i would go for 30 minutes at first to work your way up since you have not been excercising you dont want to get burned out, I go by mileage on the bike and try to at least get 2 miles in but will aim for more but it does not take long to get 2 miles in only about 6.5 minutes but i do other cardio, like walking and elliptical, but if the bike is the only resource you will use for excercise i would aim for at least 30 minutes and thats what i would start off with.
